CELLTOX – International Congress on In Vitro Toxicology

October 19-21, 2011 in Rome, Italy

In vitro toxicology has grown and acquired a major importance over the last two decades, both for the scientific advancements in this field and for the increasing influence of its approaches on the regulatory bodies. This Congress will provide a forum for presenting novel approaches and recent results and to exchange ideas among leading scientists and young researchers. IVTIP Spring 2011 Meeting

The Spring meeting of IVTIP (In Vitro Testing Industrial Platform) is in collaboration with ESTIV (European Society of Toxicology In Vitro) and CAAT (Center for Alternatives to Animal Testing). The title is “In vitro reconstructed human tissue models as alternatives to animal testing: applicability and limitations.”

AAPS Workshop on Drug Transporters in ADME

March 14-16, 2011 in Bethesda, MD

Recognition of the influence of membrane transporters on drug disposition has driven a surge in related research within the pharmaceutical sciences. Drug transport research continues to evolve rapidly, with advances in understanding the role of transport in drug absorption, distribution, and excretion, as well as toxicity and disease. Although considerable progress has been made over the past 15 years, the field of drug transport continues to evolve, particularly with respect to clinical translation of in vitro/preclinical data, understanding systemic/tissue exposure implications, toxicity/disease pathogenesis, targeting transport for drug delivery, and interplay with metabolism.

This fifth biennial AAPS Workshop aims to build on the success of previous meetings and provide an opportunity for pharmaceutical, academic and regulatory scientists to exchange ideas about transporter research.

Endocrine Disruptors 2011

AAPS – American Association of Pharmaceutical Scientists

Nov. 14-17, 2010 in New Orleans, LA

Every three to four years, the International Pharmaceutical Federation (FIP) organizes a Pharmaceutical Sciences World Congress (PSWC). Each year, the American Association of Pharmaceutical Scientists (AAPS) organizes the AAPS Annual Meeting and Exposition.

In 2010 FIP and AAPS will hold the PSWC and the Annual Meeting together in New Orleans, LA at the Morial Convention Center from November 14–18 (The expo runs from Nov. 14-17). The scientific theme is: Improving Global Health Through Advances in Pharmaceutical Sciences.

The joint meeting will include:

The keynote speaker will be Dr. Melvin Andersen, Director of the Divison of Computational Biology at The Hamner Institues for Health Sciences (formerly CIIT). Other speakers include Dr. Jim McKim of CeeTox, Dr. David Dix of the National Center for Computational Toxicology at the U.S. EPA, Dr. Michael Hornung from the U.S. EPA’s Mid-Continent Ecology Division Laboratory, and a representative from People for the Ethical Treatment of Animals.

Measuring Permeability for Poorly Soluble Drugs

Title: “A Strategy for Characterizing P-glycoprotein Substrate and Inhibitor Potential for Sparingly Soluble Drugs”

The role of efflux transporters in the potential for drug-drug interactions (DDI) is being increasingly recognized as a clinically significant phenomenon. Based on the accumulating evidence, in 2006 the FDA issued a Draft Guidance for determining whether a New Chemical Entity (NCE) is a substrate for P-glycoprotein (Pgp) efflux transport and therefore subject to DDI with other co-administered drugs which may also be substrates for the same transporter, and whether the NCE will impact the pharmacokinetics of co-administered drugs which are substrates for the same transporter (NCE as inhibitor). The general strategy for these determinations is bidirectional permeability studies in MDR1-MDCK cell monolayers.
A general requirement for conducting permeability studies is that the compound of interest has sufficient pH 7 buffer solubility in order to achieve analytically measurable concentrations of transported solute. However, many compounds of interest do not possess this level of solubility yet are effectively absorbed in vivo and successfully marketed.

Based on these considerations, we developed a method for assessing permeability of compounds with limited solubility under neutral pH conditions, utilizing Polysorbate 80 (PS80). PS80 is a very effective agent for increasing the solubility of poorly soluble compounds. However, PS80 is also a reasonably potent Pgp inhibitor in Caco-2 cells, thereby potentially masking the ability to observe solute interactions with this transporter. The present studies establish that transfected Pgp in MDCK cells is less sensitive to PS80 and support the feasibility of using such a solubilization strategy for conducting substrate and inhibitor characterizations, following the FDA recommendations for transporter mediated drug-drug interactions.
